Ces lncRNAs sont exprim\u00e9s avec une tr\u00e8s grande sp\u00e9cificit\u00e9 tissulaire et plusieurs preuves indiquent qu\u2019ils sont fonctionnellement importants, jouant un r\u00f4le r\u00e9gulateur n\u00e9cessaire au bon fonctionnement de nos cellules. Les lncRNAs sont \u00e9galement fr\u00e9quemment d\u00e9r\u00e9gl\u00e9s ou mut\u00e9s dans une grande vari\u00e9t\u00e9 de maladies humaines. Mais, leur contribution au d\u00e9veloppement et \u00e0 la progression des maladies et leur mode d&#8217;action mol\u00e9culaire restent largement inconnus. Ainsi, l\u2019un des principaux d\u00e9fis pour comprendre l\u2019influence de ces mol\u00e9cules non codantes sur la sant\u00e9 humaine est non seulement de d\u00e9terminer lesquelles sont fonctionnelles, mais aussi de d\u00e9chiffrer la fa\u00e7on dont elles accomplissent leurs t\u00e2ches et contribuent au d\u00e9veloppement de maladies.<\/p>\n<p>Notre laboratoire tire parti des donn\u00e9es g\u00e9n\u00e9tiques humaines pour identifier les lncRNAs dans les r\u00e9gions g\u00e9nomiques \u00e0 risque. Nous combinons des mod\u00e8les murins g\u00e9n\u00e9tiquement modifi\u00e9s et des mod\u00e8les cellulaires humains avec la g\u00e9nomique fonctionnelle et des techniques d\u2019\u00e9dition de g\u00e9nome bas\u00e9es sur CRISPR pour perturber les fonctions des lncRNAs et caract\u00e9riser leur r\u00f4le. Nous cherchons \u00e9galement \u00e0 d\u00e9couvrir de nouveaux m\u00e9canismes non codants bas\u00e9s sur l&#8217;ARN en caract\u00e9risant les interactions mol\u00e9culaires et les principes de r\u00e9gulation qui sous-tendent la fonction des lncRNAs. Pour cela, nous utilisons une combinaison de biochimie, d\u2019\u00e9dition du g\u00e9nome, de genomique fonctionnelle et de bioinformatique pour identifier les prot\u00e9ines interagissantes et comprendre comment des s\u00e9quences sp\u00e9cifiques m\u00e9dient la fonction des lncRNAs . Notre objectif est de mieux comprendre l\u2019impact des lncRNAs et des r\u00e9gions non codantes sur la sant\u00e9 et les maladies et de fournir les bases g\u00e9n\u00e9tiques et mol\u00e9culaires pour le d\u00e9veloppement de nouveaux diagnostics et de traitements ciblant l&#8217;ARN.<\/p>\n<\/div>\n<div class=\"one-half  \">\n<h2>Research axis<\/h2>\n<ul>\n<li>Systems Biology<\/li>\n<li>Cancer<\/li>\n<li>Genomics<\/li>\n<li>Human Genetics and Genetic Diseases<\/li>\n<li>Model Systems in Molecular Biology<\/li>\n<li>Bioinformatics<\/li>\n<\/ul>\n<h2>Research description<\/h2>\n<p>Our understanding of how the genome is regulated has changed by discovering that the vast majority of noncoding regions in the genome are transcribed, generating long RNAs that don&#8217;t encode proteins (lncRNAs). Thousands of lncRNA genes, expressed with exquisite tissue specificity, have been identified and several evidence indicate that they are functionally important and perform regulatory roles necessary for the proper functioning of our cells. lncRNAs are also frequently misregulated or mutated in a wide variety of human diseases. But, their contribution to the development and progression of diseases and their molecular mode of action remain largely unknown. Thus, one of the main challenge to understand the noncoding genome\u2019s influence on human health is not only to determine which lncRNAs are functional, but also to decipher how they perform their tasks and affect the pathophysiology of diseases.<\/p>\n<p>Our laboratory takes advantage of human genetics data to identify lncRNAs in disease risk genomic regions. We combine genetically engineered mouse models and human cell systems with functional genomics and CRISPR-based genome editing techniques to perturb lncRNA functions and characterize their role. We also aim to uncover novel noncoding RNA-based mechanisms by characterizing the molecular interactions and regulatory principles underlying the function of lncRNAs. For this, we use a combination of biochemistry, genome editing, computational and high-throughput genomics approaches to identify interacting proteins and understand how specific domains or RNA sequences within lncRNAs mediate their function.
