Nous nous int\u00e9ressons plus particuli\u00e8rement \u00e0 un groupe de g\u00e8ne\/prot\u00e9ine de la famille de Bcl-2, et \u00e0 une cascade prot\u00e9olytique gouvern\u00e9e par l&#8217;activation d&#8217;une classe de prot\u00e9ases sp\u00e9cifiques, les caspases. L&#8217;implication de certaines organelles de la cellule, dont les mitochondries et les lysosomes, dans la r\u00e9gulation de la mort des cellules tumorales est \u00e9galement sous \u00e9tude. L&#8217;influence et la r\u00e9gulation exerc\u00e9es par certaines des prot\u00e9ines de la famille de Bcl-2 sur les points-contr\u00f4le du cycle cellulaire, est \u00e9galement investigu\u00e9 dans le laboratoire, plus particuli\u00e8rement la r\u00e9gulation de Bcl-xL dans les points-contr\u00f4le G2\/M et durant la mitose.<\/p>\n<p>Des \u00e9tudes sur les m\u00e9canismes \u00e9pig\u00e9n\u00e9tiques stabilisant la s\u00e9nescence cellulaire induite par la chimioth\u00e9rapie, et sur l&#8217;influence des cycles circadiens dans les r\u00e9ponses \u00e0 la chimioth\u00e9rapie sont planifi\u00e9es dans le laboratoire.\u00a0 Finalement, de nouvelles strat\u00e9gies exp\u00e9rimentales anticanc\u00e9reuses sont \u00e9valu\u00e9es dans le laboratoire, en collaboration avec d&#8217;autres groupes de recherche.<\/p>\n<\/div>\n<div class=\"one-half  \">\n<h2>Research axis<\/h2>\n<ul>\n<li>Cancer<\/li>\n<li>Protein interactions<\/li>\n<li>Intracellular signaling<\/li>\n<li>Therapeutic approaches<\/li>\n<\/ul>\n<h2>Research description<\/h2>\n<p>The cellular and molecular responses to genotoxic drugs used in anticancer chemotherapy are extremely varied and complex. Genotoxic drugs activate a series of responses involving gene activation, post-translational protein modifications and intracellular signalling pathways that mediate cell cycle arrest, cellular senescence and\/or cell death.<\/p>\n<p>Research in the laboratory is focused on the control and function of genes, proteins and signalling pathways that regulate cell death by apoptosis, particularly on genes\/proteins of the Bcl-2 family, and on activation of a proteolytic cascade involving a class of specific proteases, the caspases.\u00a0 The importance of cellular organelles, including mitochondria and lysosomes, in the regulation of apoptosis is also investigated. The influence of Bcl-2 family members on cell-cycle checkpoints is also being investigated in the laboratory, particularly Bcl-xL regulation in the G2\/M and spindle-assembly checkpoints.<\/p>\n<p>Research projects on the epigenetic changes associated with stabilization of cellular senescence, and on the influence of circadian rhythms on tumour responses to chemotherapy are also planned.
