C\u2019est ainsi que nous avons pu r\u00e9cemment r\u00e9aliser la diff\u00e9renciation de cellules souches embryonnaires humaines en photor\u00e9cepteurs de type c\u00f4ne avec une efficacit\u00e9 encore jamais \u00e9gal\u00e9e.<\/li>\n<li><strong>BMI1 et le gliome humain<\/strong><br \/>\nLe gliome est la tumeur c\u00e9r\u00e9brale la plus fr\u00e9quente chez les personnes de plus de 40 ans. L\u2019esp\u00e9rance de vie moyenne est de 1 an. Nous avons d\u00e9couvert que le g\u00e8ne BMI1 est surexprim\u00e9 dans la fraction de cellules souches canc\u00e9reuses CD133+ et que son inactivation bloque les voies de survies AKT, FOXOA3 et P18INK4B. BMI1 joue \u00e9galement un r\u00f4le nouveau dans le recrutement de la machinerie de reconnaissance et de r\u00e9paration des dommages \u00e0 l\u2019ADN dans les cellules souches normales et canc\u00e9reuses.<\/li>\n<li><strong>\u00c9tude des m\u00e9canismes contr\u00f4lant le vieillissement neuronal<\/strong><br \/>\nLe premier facteur de risque de la maladie d\u2019Alzheimer est l\u2019\u00e2ge. La raison en demeure toujours inconnue. Nous avons d\u00e9couvert que Bmi1 joue un r\u00f4le critique dans la survie et le vieillissement des neurones en contr\u00f4lant les concentrations de radicaux libres g\u00e9n\u00e9r\u00e9s par les mitochondries. Notre laboratoire poursuit actuellement ses travaux sur les m\u00e9canismes mol\u00e9culaires de contr\u00f4le du vieillissement normal et pathologique du cerveau.<\/li>\n<\/ul>\n<\/div>\n<div class=\"one-half  \">\n<h2>Research axis<\/h2>\n<ul>\n<li>Development and tissue differentiation<\/li>\n<li>Stem cells<\/li>\n<li>Neurobiology<\/li>\n<li>Model systems in molecular biology<\/li>\n<\/ul>\n<h2>Research Description<\/h2>\n<ul>\n<li><strong>Genetics of Retinal and Stem Cell Development<\/strong><br \/>\nOur work in this area has allowed us to integrate our fundamental knowledge of the molecular mechanisms controlling neurogenesis and retinogenesis with stem cell biology. This is how we were recently able to differentiate human embryonic stem cells into cone-like photoreceptors with unprecedented efficiency.<\/li>\n<li><strong>BMI1 and Human Glioma<\/strong><br \/>\nGlioma is the most common brain tumor in people over 40. The average life expectancy is one year. We discovered that the BMI1 gene is overexpressed in the CD133+ cancer stem cell fraction and that its inactivation blocks the AKT, FOXOA3, and P18INK4B survival pathways. BMI1 also plays a novel role in recruiting DNA damage recognition and repair machinery in normal and cancerous stem cells.<\/li>\n<li><strong>Study of the Mechanisms Controlling Neuronal Aging<\/strong><br \/>\nThe primary risk factor for Alzheimer&#8217;s disease is age. The reason for this remains unknown. We have discovered that Bmi1 plays a critical role in neuronal survival and aging by controlling the concentrations of free radicals generated by mitochondria.
