D&#8217;un point de vue clinique, des mutations de nombreux g\u00e8nes du syst\u00e8me ubiquitine ont \u00e9t\u00e9 li\u00e9es de mani\u00e8re causale \u00e0 diverses maladies, dont le cancer. L&#8217;ubiquitination est un processus r\u00e9versible d\u00fb \u00e0 la pr\u00e9sence des d\u00e9ubiquitinases (enzymes qui enl\u00e8vent sp\u00e9cifiquement l&#8217;ubiquitine des prot\u00e9ines cibles). Bien que les d\u00e9ubiquitinases soient peu connues, elles sont n\u00e9anmoins en train d&#8217;\u00e9merger comme des r\u00e9gulateurs centraux du syst\u00e8me ubiquitine.<\/p>\n<p>Notre objectif est de caract\u00e9riser les fonctions biologiques et les m\u00e9canismes d&#8217;action des d\u00e9ubiquitinases impliqu\u00e9es dans la transcription et la r\u00e9paration de l&#8217;ADN, deux processus dont la d\u00e9r\u00e9gulation joue un r\u00f4le fondamental dans le d\u00e9veloppement du cancer.<\/p>\n<p>Nous utilisons des approches de biochimie, de biologie mol\u00e9culaire et de culture cellulaire, \u00e0 la fine pointe de la technologie, afin de caract\u00e9riser les fonctions des d\u00e9ubiquitinases.<\/p>\n<p>Nos travaux permettent une meilleure compr\u00e9hension des m\u00e9canismes mol\u00e9culaires gouvernant le syst\u00e8me ubiquitine. De plus, de nouveaux concepts pourront \u00eatre d\u00e9velopp\u00e9s concernant la r\u00e9gulation de la transcription et la r\u00e9paration de l&#8217;ADN. Clinically speaking, the mutations of many genes in the ubiquitin system have been causally linked to various diseases, including cancer. \u00a0Ubiquitination is a reversible process due to the presence of deubiquitinating enzymes, or enzymes that specifically remove ubiquitin from target proteins. Although deubiquitinating enzymes are not well understood, they are nevertheless emerging as central regulators in the ubiquitin system.<\/p>\n<p>Our goal is to characterize the biological functions and mechanisms of action of the deubiquitinating enzymes involved in DNA transcription and repair, two processes whose deregulation plays a key role in the development of cancer.<\/p>\n<p>We use state-of-the-art biochemistry, molecular biology, and cell culture approaches to characterize the functions of deubiquitinating enzymes. Our work provides a better understanding of the molecular mechanisms governing the ubiquitin system.
